INVESTIGATING THE ROLE OF HIPPOCAMPUS - ORBITOFRONTAL CIRCUITS FOR COGNITIVE FLEXIBILITY

US · IL NIH grant awarded #nih-5R01MH126105-05

Summary

This project investigates the neural circuits involving the hippocampus and orbitofrontal cortex that underlie cognitive flexibility and stress resilience, with the goal of identifying new therapeutic targets for psychiatric disorders.

What they want

The project will study the role of ventral hippocampus-orbitofrontal cortex (OFC) circuits in reversal learning and stress resilience. Aim 1 will inhibit specific neural projections within this circuit (ventral hippocampus to medial OFC, and medial OFC to lateral OFC) to test its functional importance for reversal learning. Aim 2 will use in vivo Ca2+ imaging to examine neural activity in the ventral hippocampus, medial OFC, and lateral OFC to understand how neurons store, process, and update action-outcome value associations for reversal learning. Aim 3 will investigate how these brain regions become dysfunctional under chronic stress and test if stimulating this circuitry can confer stress resilience and counteract stress-induced deficits in reversal learning.
Deliverables
  • First insight into a new element of the neural circuitry underlying cognitive flexibility and stress resilience
  • Identification of potential new neural circuit-based targets for novel drugs or advanced cognitive-behavioral therapies aimed at improving cognitive flexibility in patients suffering from psychiatric disorders
Technical requirements
  • Inhibition of direct input projections from ventral hippocampus to medial OFC
  • Inhibition of output projections from medial OFC to lateral OFC
  • In vivo Ca2+ imaging of neural activity in ventral hippocampus, medial OFC, and lateral OFC
  • Chronic stress models
  • Stimulation of neural circuitry
