Summary

This project investigates the continuous nature of epidermal differentiation, focusing on the gene-regulatory program that maintains basal-to-spinous transition cells, a recently recognized cell state.

What they want

The research will utilize single-cell and spatial transcriptomics to analyze cellular composition, gene expression, and cell-cell signaling in P0 epidermis from mice with knockout transcription factors. It will also employ single-nuclear ATAC-seq with single-nuclear RNA-seq to map changes in open chromatin and define transcription factor binding and histone modifications in sorted epidermal cells. Computational modeling will be used to define underlying gene regulatory networks and inform functional testing of signaling pathways and transcription factors.
