Summary

This project proposes to continue systematic analysis of complex genetic interaction networks and phenotypes in yeast and human cells to understand gene function and identify disease gene modifiers.

What they want

The project will perform systematic phenotypic and genetic analyses in wild, genetically diverse yeast strains to identify genetic modifiers underlying background-specific gene essentiality and map genetic interactions. It will also produce quantitative cell biological phenotypic profiles associated with gene perturbation, exploring the influence of cell state on genetic perturbation effects using proteome dynamics. Finally, it aims to map a global genetic interaction network for a human cell line by screening an informative set of query gene mutants, with an emphasis on essential genes, to expand the scaffold genetic network and map networks underlying functionally representative protein complexes.
